The van, which was carrying three prisoners, flipped on to its side on the northbound sliproad to the Winchester Services on the M3 shortly before 4.50pm.
The driver of the vehicle, which belongs to Reliance Security Group, sustained serious injuries which were described as non-life threatening.
Police and ambulance crews are still on scene attempting to free the driver from the van.
Hampshire and Isle of Wight Air Ambulance has also been called to attend the incident.
A spokeswoman for Hampshire Police said: "All three prisoners are accounted for, they are complying with officers and none of them are injured.
"The driver is currently trapped in the vehicle with serious but non-life threatening injuries.
"No-one else was injured in the incident."
It is not yet known where the vehicle had been travelling to and from, or how the accident occurred.
The incident has led to chaos with traffic at a standstill on the motorway.
