Jimmy Ball's Stags were edged out 1-0 by National League South high-fliers Havant & Waterlooville.
Muhammadu Faal netted the only goal, capitalising on a defensive mix-up to turn in.
Lewis Noice denied Jason Prior doubling the lead before half-time, before Totton improved after the restart.
However, they could not find a way through, and saw a late penalty shout waved away, as Havant progressed.
Sholing were also beaten by a side two tiers higher, going down 3-0 at Chippenham Town.
Jordan Young put the Bluebirds in front on 36 minutes, before Dave Diaper's Boatmen felt they should have been awarded a penalty when Dan Mason was brought down.
Instead, a free-kick was awarded just outside the box, with goalkeeper Will Henry only shown a yellow card.
Chippenham struck twice more in the final 20 minutes, through Luke Russe and Harvey Greenslade to seal their spot in Tuesday's draw.
